Description
LOWENSTEIN JENSEN MEDIUM is a medium used for the cultivation of mycobacteria, in particular Mycobacterium tuberculosis, whether from clinical sources (sputum, gastric lavage) or pure culture.
The medium is in accordance with the formulation described by Lowenstein, modified by Jensen which eliminated the use of congo red, increasing the concentration of malachite green. In accordance with international guidelines (NHS - Standards Unit, Evaluations and Standards Laboratory Centre for Infections), samples should be pre-treated before inoculation to control and reduce contamination if present.
No surveys or ratings are available to recommend an optimal method. The choice of the appropriate method and the time of action vary depending on the degree of contamination of the samples. The L.J. Medium can be modified (Lowenstein Jensen-Gruft) by the use of antibiotics such as, penicillin (50 U/ml), nalidixic acid (35 mcg/ml) and RNA (80 mcg/ml), since they inhibit concomitant microbial flora and RNA further favors the development of Mycobacterium tuberculosis.
