Description

Kligler Iron Agar (KIA) is a medium used for Enterobacteriacae differentiation (confirmatory test), based on their ability to ferment sugars, such as glucose and lactose, and to produce hydrogen sulfide.

Sugar fermentation is detected by the medium's yellowing, while the production of H2S is detected by the medium's blackening. The phenol red is present as a pH indicator, instead ammonium citrate iron is present as an indicator of H2S.

The KIA has an identical TSI medium composition, with the exception that the TSI contains, in addition to glucose and lactose, also sucrose. The KIA agar is arranged to be on sloped; with the upper portion exposed to oxygen (aerobic chamber), while the lower portion (bottom) is oxygen-free (anaerobic chamber).
